James Moncrief Brailsford, Jr. was born on July 3, 1919 in Orangeburg, SC, the son of James Moncrief Brailsford and Mary Elizabeth "Bessie" (Bates) Brailsford. He graduated from the University of South Carolina in 1934.

After passing the SC bar, James Moncrief Brailsford, Jr. launched his own law firm in Orangeburg, SC.

James Moncrief Brailsford, Jr. married Louise Rook Tompkins, daughter of Frank Tompkins and Martha (Aldrich) Tompkins of Columbia, SC; they had two known daughters and two known sons. He married a second time, to Joan Ward Culler.

In 1938, James Moncrief Brailsford, Jr. was first elected as one of five men to represent Orangeburg County in the SC House of Representatives of the:
- 83rd General Assembly that met from 1939 to 1940
- 84th General Assembly that met from 1941 to 1942

On November 30, 1943, James Moncrief Brailsford, Jr. enlisted in the U.S. Army at Fort Jackson, SC and served during World War II.

In 1946, James Moncrief Brailsford, Jr. was again elected as one of four men to represent Orangeburg County in the SC House of Representatives of the:
- 87th General Assembly that met from 1947 to 1948
- 88th General Assembly that met from 1949 to 1950

On January 26, 1949, James Moncrief Brailsford, Jr. was elected a Judge of the 1st Judicial Circuit, and he served until he was elected to the SC Supreme Court. He resigned from the House of Representatives on July 2, 1949.

On March 6, 1962, James Moncrief Brailsford, Jr. was elected as an Associate Justice on the SC Supreme Court, replacing Associate Justice George Dewey Oxner, who had recently died. He continued as an Associate Justice on the SC Supreme Court until December 5, 1974, when he reached the mandatory retirement age.

On November 8, 1993, James Moncrief Brailsford, Jr. died, and he was buried in the Sunnyside Cemetery in Orangeburg, SC.
