Thomas Boone Fraser, Jr. was born on June 21, 1860 in Sumter, SC, the son of Thomas Boone Fraser, Sr. and Sarah Margaret (McIver) Fraser. He first attended private schools. In 1876, he entered Davidson College in North Carolina, and he graduated with an A.B. in 1881. He then studied law under his father, Judge Thomas Boone Fraser, Sr. of the 3rd District Court. He was admitted to the SC bar in 1883.

On December 16, 1886, Thomas Boone Fraser, Jr. married Emma Marshall Edmunds, daughter of Nicholas William Edmund and Mary Claudia (Leland) Edmund; they had one known daughter.

In 1900, Thomas Boone Fraser, Jr. was first elected as one of five men (one of three men after 1902) to represent Sumter County in the SC House of Representatives of the:
- 64th General Assembly that met from 1901 to 1902
- 65th General Assembly that met from 1903 to 1904
- 66th General Assembly that met from 1905 to 1906
- 67th General Assembly that met from 1907 to 1908
- 68th General Assembly that met from 1909 to 1910
- 69th General Assembly that met from 1911 to 1912

On January 11, 1912, Thomas Boone Fraser, Jr. was elected as an Associate Justice on the SC Supreme Court, replacing Eugene Blackburn Gary who had been elected Chief Justice. He resigned from the SC House of Representatives on the same date as his election to the SC Supreme Court. He was an Associate Justice on the SC Supreme Court until his death.

On May 21, 1925, Thomas Boone Fraser, Jr. died, and he was buried at the Sumter Cemetery in Sumter, SC.
